My research aims at the enhancement of biochemical properties, the gain of function and the improved understanding of oxidoreductases. Hereby, the flavin containing enzyme pyranose 2-oxidase (P2O, POx) serves as a model enzyme to be used as the basis for the development of a novel ultra-high-throughput screening. This screening makes use of genetically encoded, fluorescent sensors derived from the green fluorescent protein (GFP) and allows using fluorescent activated cell sorting (FACS) as a screening tool. Furthermore, a key topic and ultimate goal of my research is to test the screening platform in a directed evolution experiment performed on the P2O enzyme.
